Taking a less-senior role in order to gain skills? Is this possible/wise?

I'm a Analytical Chemist working in pharma and looking to broaden my skill set to include additional analytical techniques (mass spectrometry in particular). My current role does not allow for me to explore and broaden my skill set and my managers have explicitly denied me the chance at working on other teams to gain these skills.

Along with horrible work-life balance (company culture has shifted) I'm looking to leave and find a role where I can broaden said skillset. The issue is that I'm mid-senior/director level at this point and any jobs that would allow me to learn MS also require that I'm already fluent in the technique. The only jobs I might actually qualify for are associate level, which I would be overqualified for based on experience and education (I have a PhD and entered the workforce at mid-senior).

Money and status are not a concern of mine and I would gladly take a paycut and a demotion if it meant gaining new skills. Am I being naive? Or is it possible to take a step back and re-align my career path?
