Should I call the company before applying?

I'm about to apply for a helpdesk job, having just graduated. My dad, a manager, said my cover letter was flawless, but that I should call up the company and ask to speak to the hiring manager to get their name and make the cover letter more personal (use their name instead of Dear Hiring Manager), and so that my foot is in the door early. He's suggested I ask a couple of questions (though I don't really have any I want to ask; they'd be ones I come up with for the sake of it).

I'm of the mind that the phone call could go either way, and that it could show I don't follow instructions (there isn't a number to call on the ad, and I'd be going through the receptionist to ask for the hiring manager). I could also blow the phone call and give a bad first impression before they read my application.
