If a job ad has a contact number and email for the hiring manager, what should I say when I call them?

Apologies if this has already been posted . When I have done this in the past it's been more just to see how many years of experience they want. Now my mentality is I can speak directly to the hiring and line manager so I should take advantage and present myself in a good light before I even apply. Also, should I email the application to her as well as applying through the website?
