Writing a Note of Interest for a Promotion

Hello, a promotion opportunity has opened up at my work that I'd like to apply for. The advert says I need to write a note of interest and I'm not quite sure what that entails, nor how to write one.

I'd be getting promoted from Customer Assistant to Team Support.

The add itself calls for people who are good with problem solving as well as good people skills - I have both, but I don't know how to communicate that.

And lastly, once the note is written, how do I hand it to my manager? (presentation wise like in an envelope etc)
