To follow up or not? Interview request

I sent in a cover letter 3 days ago for a management position at 1pm. I received a response at 3pm "at what point are you available for an interview?". I replied at 4pm, thanking him for getting back to me and giving him my schedule for the next week when i was available. I still have not receieved a reply to my response. Should I send a follow up in case it my reply got "lost", saying i am very interested but understand the selection process shouldnt be rushed, thanking him again, and casually letting him know my work schedule over the coming weeks?

I dont want to seem desperate, but I also am eager for the opportunity. Im happy at my current job (asst manager) so I really have nothing to lose, and everything to gain.
